Desensitization — how fast does it actually happen on GHRPs
18 posts
Everyone says 'cycle off or your receptors downregulate.' The question I want to actually pin down: how fast, and how measurable?
What I've pieced together:
- Ipamorelin: pulse amplitude starts declining around week 4, noticeable decline by week 6, half-amplitude by week 8 at steady dose.
- Hexarelin: faster — week 2-3 is the typical knee.
- GHRP-2/GHRP-6: somewhere in between.
- GHRH analogs (CJC, mod-GRF): slower desensitization, more about pituitary depletion than receptor downreg.
Recovery after washout: 3-4 weeks typically restores full sensitivity.
Strategies:
- 5-on / 2-off weekly — works for some, anecdotal
- Hard 8-on / 4-off — standard
- Dose escalation to compensate — makes the problem worse
- Compound rotation (ipa → hexarelin → GHRP-6) mid-cycle
How are people measuring desensitization in real time? Blood IGF? Subjective recovery? Sleep depth?

Subjective marker that's held up: 'morning fasted hunger.' On GHRP the ghrelin-receptor agonism gives a hunger bump post-pin. When desensitization kicks in, that hunger bump fades. I use it as a free crude biomarker.

5-on/2-off has held up for me across 6+ cycles of longer duration. Not proof but the subjective-and-wearable curve is flatter than on straight 7 days/wk. Feels like it extends the useful window to ~10-12 weeks.

The GH secretagogue receptor downregulation timeline in humans is not well characterized. Most of what we 'know' comes from a handful of studies in GH deficiency patients plus anecdotal forum reports. Be careful about precise claims.

Compound rotation mid-cycle is a real strategy. I know vets running ipa weeks 1-3, hexarelin weeks 4-5, GHRP-6 weeks 6-8. Each one hits a slightly different receptor avidity profile, extending total window. Not for beginners.

Dose escalation to 'push through' desensitization is the worst of the options. You get mild extension and harder sides. Cycle off wins.

The 3-4 week recovery number I'd quibble with slightly. For Ipa that holds. For hexarelin, sensitivity recovery is more like 5-6 weeks in my experience.

Great data points. Consolidating: IGF-1 at 2/4/6/8, subjective hunger pulse post-pin, wearable deep sleep trend. Three cheap markers, probably sufficient. Compound rotation is next-level but real.

Saving. This thread is the best summary of desensitization markers I've read.

Would love a community-assembled dataset of IGF curves across cycles. Could probably identify responder archetypes.
